Activity of liver microsomal enzymes during the chronic phase of murine schistosomiasis BJMBR
Conte,F.P.; Fidalgo-Neto,A.A.; Manhães-Rocha,D.A.; Paumgartten,F.J.R.; De-Oliveira,A.C.A.X..
The effects of schistosomiasis on microsomal enzymes were studied on post-infection day 90 when accumulated damage and fibrosis are most intense but granulomatous reaction around the eggs harbored in the liver is smaller than during the earlier phases. Swiss Webster (SW) and DBA/2 mice of either sex (N = 12 per sex per group) were infected with 100 Schistosoma mansoni cercariae on postnatal day 10 and killed on post-infection day 90. Cytochrome P-450 (CYP) concentration and alkoxyresorufin-O-dealkylases (EROD, MROD, BROD, and PROD), p-nitrophenol-hydroxylase (PNPH), coumarin-7-hydroxylase (COH), and UDP-glucuronosyltransferase (UGT) activities were measured in hepatic microsomes. Age-matched mice of the same sex and strain were used as controls. In S....

Collagen arrangement in hepatic granuloma in mice infected with Schistosoma mansoni: dependence on fiber radiation centers BJMBR
Lenzi,H.L.; Kimmel,E.; Schechtman,H.; Pelajo-Machado,M.; Vale,B.S.; Panasco,M.S.; Lenzi,J.A..
The collagen structure of isolated and in situ liver granuloma from Swiss Webster mice infected with Schistosoma mansoni was sequentially and three-dimensionally analyzed during different times of infection (early acute, acute, transitional acute-chronic, and chronic phases) by laser scanning confocal microscopy and electron scanning variable vacuum microscopy. The initial granuloma structure is characterized by vascular collagen residues and by anchorage points (or fiber radiation centers), from where collagenous fibers are angularly shed and self-assembled. During the exudative-productive stage, the self-assembly of these fibers minimizes energy and mass through continuous tension and focal compression. The curvature or angles between collagen fibers...

Distinct nonrandom patterns of chromosomal deletions in giant-cell lesions of bone Genet. Mol. Biol.
Baruffi,Marcelo Razera; Barbieri-Neto,J.; Pina-Neto,João Monteiro; Suerzut,C.E.; Casartelli,Cacilda.
Cytogenetic analyses were performed on a bone giant cell reparative granuloma (GCRG) and on three bone giant cell tumors (GCT). The present GCRG case is the second to be described cytogenetically. A modal chromosome number of 46 was observed in all samples. Clonal chromosome abnormalities were detected in all cases. The numerical alterations most frequently observed involved the loss of chromosomes 17 and 18. Among the structural anomalies observed, there was preferential involvement of chromosomes 6 and 10. Three GCT cases presented del(10)(p13) and two cases presented del(6)(q25) (1 GCRG and 1 GCT). These breakpoints mapped on 10p and 6q may harbour genes of importance in the development of bone giant cell tumors.

BUSTAMANTE,JAVIER ANDRÉS; ASTUDILLO,MIRYAM; PAZOS,ALVARO JAIRO; BRAVO,LUIS EDUARDO.

Indirect effects of oral tolerance to ovalbumin interfere with the immune responses triggered by Schistosoma mansoni eggs BJMBR
Carvalho,C.R.; Lenzi,H.L.; Correa-Oliveira,R.; Vaz,N.M..
The objective of the present study was to investigate whether the injection of a tolerated protein (indirect effects) affects the formation of granulomas around Schistosoma mansoni eggs trapped in the lungs after intravenous (iv) injection into normal (noninfected) C57BL/6 mice (6 animals per group). To induce oral tolerance to chicken egg ovalbumin a 1/5 dilution of egg white in water was offered ad libitum in a drinking bottle for 3 days. Control mice received water. After 7 days, control and experimental animals were injected iv with 2,000 S. mansoni eggs through a tail vein. In some mice of both groups the iv injection of eggs was immediately followed by intraperitoneal (ip) immunization with 10 µg of dinitrophenylated conjugates of ovalbumin (DNP-Ova)...

Intra-abdominal granulomas caused by Clostridium tertium in an American Fuzzy Lop rabbit Ciência Rural
D’Elia,Mirella Lauria; Santos,Alice Barroso; Ribeiro,Beatriz Novaes Telles; Torres,Renato Cesar Sacchetto; Santos,Renato de Lima; Silva,Rodrigo Otávio Silveira; Nepomuceno,Anelise Carvalho.
ABSTRACT: A 6-year-old Fuzzy Lop rabbit was referred to a veterinary hospital with a complaint of lameness. In addition to a vertebral subluxation, two radiopaque and well-defined structures were revealed by radiographic evaluation. Ultrasonographically, the masses were characterized as parenchymal structures with diffuse mineralization and formation of reverberation artifacts, suggesting presence of gas. These two structures were excised during a laparotomy and Clostridium tertium was isolated. To the best of our knowledge, this is the first report of C. tertium infection in a pet animal.

Morphometric Analysis of Granulomas Induced by Mycobacterium bovis Suggests an Influence of IFN-Gamma on the Generation and Modulation upon Granulomatous Inflammatory Response in the Different Tissues International Journal of Morphology
Azevedo Duarte,Tonya; Almeida Barbosa Jr.,Aryon de; Arruda,Sergio.
There is evidence in both human and experimental infection caused by Mycobacterium tuberculosis, that immunologic factors influence susceptibility to infection and the progress of the disease. The present study aims to evaluate the role of IFN- g upon inflammatory granulomatous response against M. bovis. To pursue that, C57BL/6 mice lacking the genes for synthesis of IFN- g (IFN- g-/-) and their wild-type counterparts (IFN- g+/+) were intravenously inoculated with M. bovis. The ability of M. bovis to survive and replicate in the liver and lungs was evaluated by counting colony-forming unit (CFU). The histopathological features of granulomatous inflammatory response in the liver and lungs were analyzed during the infection by M. bovis. Granuloma parameters...

Mycobacterium marinum infection: a case report J. Venom. Anim. Toxins incl. Trop. Dis.
Sette,Christiane Salgado; Wachholz,Patrick Alexander; Masuda,Paula Yoshiko; Figueira,Renata Borges Fortes da Costa; Mattar,Fernanda Rodrigues de Oliveira; Ura,Deise Godoy.
AbstractThe infection by Mycobacterium marinum in humans is relatively uncommon. When it occurs, it mainly affects the skin, usually with a chronic, indolent and benign evolution. The diagnosis requires a high index of suspicion, and a significant delay may be observed between the first symptoms to the final diagnosis. This present case reports a M. marinum infection in an immunocompetent patient that had a chronic undiagnosed injury on the dominant hand for at least five years. The patient had several medical consultations, without proper suspicion, hampering adequate diagnostic investigation. Histopathology detected tuberculoid granulomas, but showed no acid-fast bacilli. The culture in appropriate medium and the polymerase chain reaction-restriction...

Revisiting the Rich's formula: an update about granulomas in human tuberculosis BJID
Coelho Filho,João Carlos; Takenami,Iukary; Arruda,Sérgio.
The formula proposed by Rich in 1951 explained the formation in a tuberculous lesion in a period that was unknown cellular functions, cytokines and other immunological aspects involved in granuloma formation by tuberculosis; its components are assembled conceptually to explain the pathogenic mechanisms involved in the granulomatous lesion in tuberculosis. In this manuscript, we report an update of Rich's formula based on the new and old concepts about pathogenic mechanisms involved in the granulomatous lesion in tuberculosis. Current knowledge allows us to conclude that the balance between the characteristics of the bacillus and host protective response is necessary to indicate the outcome of pathogenesis, infection or active disease and the necrosis...

Stem Cells and Androgen Receptor in Human Periapical Periodontitis International Journal of Morphology
Rodríguez,H; Espinoza-Navarro,O; Peñafiel,C; Escobar,E; Silva,I; Quiroz,P; Arriaza,C; Castro,M. E; Sánchez,C; Arias,M; Michea,A.
Activation of macrophages in periapical granulomas occurs through the presence of cytokines, endotoxin and other genetic and epigenetic factors, allowing the initiation of inflammation and bone resorption. The present study aims to analyze the presence of CD133 protein (marker of stem cells) and the AR (androgen receptor) protein in biopsies of human odontogenic periapical granuloma. Biopsies from 14 adult male patients with diagnosis of periapical granuloma included in paraffin blocks were processed histologically to obtain 5-µm thick sections. Protein presence was detected and analyzed by immunohistochemistry of CD133 and AR. The quantification considered the number of positive cells in 0.17 mm2 random areas under the microscope using a 1000X objective....
